Heterologous biosynthesis is a methodology to access valuable small molecule natural products via genetic transfer of the responsible gene cluster from a native producer to an enabling surrogate host.  In particular, our group uses Escherichia coli for pathway reconstitution. In one case to be presented, erythromycin, a potent polyketide antibiotic with a modular and complex biosynthetic pathway, was produced.  As such, the new system is poised for cellular engineering to redirect and optimize erythromycin formation. In a separate example, yersiniabactin, a mixed nonribosomal peptide-polyketide siderophore, was generated and the resulting system used for novel production purposes and applications that highlight the metal chelating nature of the compound.
